Development of dormancy induction in micropropagated broad leaf trees for long time storage in vitro (OG: DENDROTEC)
Rooted as well as unrooted shoot cultures in vitro kept on sterile culture media can be stored at low temperatures over month. But micropropagated cuttings of broad leaf trees rooted in sterile miniplugs directly cannot do this without vitality loss. On the other hand dormant plantlets of broad leaf trees (winter dormancy) are often stored at low temperatures as they do this in winter time. The main goal of this project will be induction of dormancy of plantlets grown in miniplugs under sterile conditions. Storage of these dormant plantlets will give a continuous production chain independent from the growing season.
The experiments on the induction of dormancy have shown that neither long-term culture at temperatures above 20°C, even under reduced day length (short day), nor senescence-inducing hormone treatment (ABA) can induce dormancy. Senescence and dormancy can only be induced by cold treatment under light exposure. Storage for up to 25 weeks was possible so that the plants could be brought into the greenhouse in early summer. At the end of storage, dead leaves were removed and the trays were kept warm in the greenhouse. New shoots appeared after approx. 1 week.
